BPC-157: Three Decades of Croatian Research

A 2003 study in a transected rat Achilles tendon model reported increased load to failure, higher Young’s modulus of elasticity, and higher Achilles functional index scores in treated animals against controls, with histological differences in fibroblast, reticulin, and collagen formation. A 2014 study in cultured tendon fibroblasts examined growth hormone receptor expression, offering a proposed mechanism for the tendon results. A 2010 study extended the model set to medial collateral ligament transection in rats, and a 2006 study to complete quadriceps transection.

That volume is not accidental. BPC-157 has been investigated since the 1990s by Predrag Sikiric and colleagues at the School of Medicine, University of Zagreb, in one of the more sustained single-compound programmes in the peptide literature. Their 2016 review in Current Neuropharmacology sets out the brain-gut framing that organises much of the work, and describes the peptide as stable in human gastric juice, a property that shaped how it has been administered across studies.

The compound is a synthetic 15-residue peptide, sequence GEPPPGKPADDAGLV, molecular weight 1419, corresponding to a partial sequence of a protein identified in human gastric juice.

The model species matters when reading any of it. The overwhelming majority of published BPC-157 studies are rodent work, and BPC-157 holds no approved human indication in any major jurisdiction.
